Expression of neuronal nuclear antigen and neuron-specific enolase in the developing spinal cord of human embryos

摘要:

目的 探讨神经元核心抗原(NeuN)和神经元特异性烯醇化酶(NSE)在人胚胎脊髓发育阶段的分布规律及其表达意义。 方法 应用免疫组织化学法检测第2、3、4个月龄段,共16例人胚胎,脊髓前角、后角中NeuN和NSE的表达、分布状况。 结果 在脊髓后角处:第2个月胚龄段,NeuN呈阴性表达, 部分细胞呈NSE阳性表达;第3个月胎龄段,部分细胞开始呈NeuN阳性表达,NSE则呈广泛阳性表达;第4个月胎龄段,NeuN阳性细胞表达数量和强度均较前增高,NSE阳性表达与第3个月龄相似。在人胚胎脊髓前角处: 第2~4个月龄段,NeuN和NSE呈广泛阳性表达。 结论 人胚胎脊髓发育第2~4个月龄段,NeuN和NSE阳性表达在脊髓的前角处早于后角,NeuN和NSE参与脊髓神经元的发育成熟过程。

Abstract:

Objective To investigate the neuronal nuclear antigen(NeuN) and neuron-specific enolase (NSE) distribution in the developing stage of the human embryonic spinal cord and their clinical significance. Methods Sixteen cases of human embryos, from 2 to 4 month-age were used in the present study. Immunohistochemical staining was uesd to detect the expression and distribution of NeuN and NSE in the anterior and posterior horn of the spinal cord.
Results At the second month of gestation, NeuN negative expression in posterior horn and NSE positive expression in some cells of the posterior horn were observed. At the third months, NeuN positive expression was first in some cells, and NSE was widely expressed in the posterior horn. At the fourth month, the number and intensity of NeuN positive expression were higher than before, and NSE positive expression was similar to that at the third month. From the second to fourth month of gestation, NeuN and NSE positive expressions were extensive in the anterior horn of human embryonic spinal cord.
Conclusion NeuN and NSE positive expressions are appeared earlier in the anterior horn than posterior horn. From the second to fourth month of gestation, NeuN and NSE may regulate neurons in the mature process during the growth and development of human embryonic spinal cord.
